Mitsui was born on March 30, 1944 in Tokyo, Japan; the son of Tametomo and Miwako (Nakayama) Mitsui.

Mitsui received a Bachelor of Arts degree from the University of Tokyo in 1967. Two years later he was given a Master of Science degree and a Doctor of Science degree from Kyoto University in 1981.
Mitsui began his career as a research associate at the Insitute for Mathematical Sciences of Kyoto University in 1969. Fourteen years later he held the position of an assistant professor and associate professor at Fukui University. Then in 1986, Taketomo took a position of a professor at Nagoya University.
In 1992, he was appointed a professor at Graduate School of Human Informatics of Nagoya University. Also Mitsui worked as a professor of Graduate School of Information Science of Nagoya University from 2003 to March 2007. In 2008, he became a professor of Department of Mathematical Sciences at Doshisha University and held the position for six years.
Mitsui is a member of Japan Society for Industrial and Applied Mathematics, Japan Mathematical Society and American Society for Industrial and Applied Mathematics.
On October 10, 1970 Taketomo Mitsui married Shoko Takizawa. They have 2 children.
