Holmes began his career as a faculty member at Howard University in 1932 and had held it for thirty-eight years. In 1961, he was appointed a professor of philosophy at the same university, where he worked until 1970. Also in 1952, Eugene became a chairman of the philosophy department at Howard University.
Holmes was a member of the Washington Philosophical Society, American Philosophical Association and Philosophical Society of Washington.
On January 2, 1939 Eugene Holmes married Margaret Cardozo.
