Maryan Ainsworth was born on September 2, 1949, in El Dorado, Kansas, United States. She is a daughter of John Charles Wynn and Rachel (Linnell) Wynn.
In 1972, Maryan received a Bachelor of Arts degree from Oberlin College. The following year, in 1973, she got a Master of Arts degree from the same educational establishment. Later, Ainsworth enrolled at Yale University, graduating with a Master of Philosophy degree in 1976 and a Doctor of Philosophy degree in 1982.
In 1977, Maryan began her career at the Metropolitan Museum of Art in New York City as a research associate, a post she held till 1980. Also, since 1977, she has been serving as a curator of European paintings at the museum. Between 1981 and 1982, she served as a research investigator. In 1982, Ainsworth was appointed a senior research associate and she remained in this position till 1987, when she was made a research fellow. In 1992, Maryan was promoted to the post of a senior research fellow at the same museum.
Also, between 1986 and 1991, Ainsworth acted as an adjunct associate professor at Barnard College. In 1991, she began working as an adjunct professor of Art History at both Barnard College and Columbia University, where she still works. The same year, in 1991, Maryan was appointed a guest scholar at J. Paul Getty Museum.
In 2018, Maryan was appointed a Kress-Beinecke Professor at the Center for Advanced Study in the Visual Arts (CASVA) at the National Gallery of Art in Washington, D.C.
In addition, during her career, Ainsworth served as a member of board of directors of Belgian-American Educational Foundation.

Maryan married Charles Herbert Ainsworth on June 5, 1971. Their marriage produced one child - Clark Emerson Ainsworth.
