Totir was born on May 1, 1936, in Bala, Romania; the son of Dumitru Ionita and Nicolita Petre Totir.

Totir received a Bachelor of Arts degree from the University of Bucharest in 1961. Fourteen years later he was given a Doctor of Philosophy degree from Institute of Physical Chemistry of Romanian Academy.
Totir began his career as a research chemist at Onesti Chemical Factory in 1961. Three years later he held the same position at the Institute of Physical Chemistry in Bucharest, Romania. Then he moved to the United states and in 1969, Nicolae became a research associate at the University of Pennsylvania in Philadelphia.
Since 1971 Totir was a senior researcher at the Institute of Physical Chemistry. He worked as an assistant professor at Politehnica University in Bucharest from 1979 to 1994. In 2001, Nicolae was appointed a professor at Pitesti University and held it for a year. Nowadays he is a Professor Emeritus at the Institute of Physical Chemistry.
Totir was a member of Romanian Chemical Society and International Society of Electrochemistry.
Nicolae Totir was married to Lucica Nicolae Gujba. They have 2 children.
