Carl Holsøe was born on March 12, 1863 in Aarhus, Denmark, into the family of an architect N.P.C. Holsøe. He also had a younger brother Niels Holsøe who was also a painter.

Carl studied at the Royal Danish Academy of Fine Arts in Copenhagen during 1882 – 1884 and then at the Kunstnernes Frie Studieskoler under Peder Severin Krøyer.
Carl made his début as a painter at the 1886 Charlottenborg December Exhibition with his painting "Interior", which was described as "having almost the character of a manifesto." From 1888 to 1910 and from 1922 to 1933 he showed his works in the Charlottenborg Spring Exhibition, and in 1909 and 1914 in the Autumn Exhibition. He also participated in the Association for National Art Exhibitions and was a member of the board of the Kunstforeningen from 1902 to 1905 and from 1909 to 1911. Holsøe's paintings include landscapes, still lifes and flowers, but he primarily painted domestic interior scenes. Carl died on November 7, 1935 in Asserbo.

In 1894 he married Emilie Heise, who was his most frequent model, but she died in 1930. On October 21, 1935, shortly before his death, he married Ingeborg Margrethe Knudsen.
