Charles Henry Burnett was born on May 28, 1842 in Philadelphia to Eli Seal Burnett and Hannah Kennedy Mustin Burnett.
He received his preliminary education in Philadelphia. He graduated from Yale College in 1864 and from the Medical Department of the University of Pennsylvania in 1867.
In the course of his studies abroad he had become much interested in diseases of the ear, and in 1870 he gave up his practice and went to Vienna to pursue his otological studies under Politzer and to Berlin where he worked with Virchow and Helmholtz.
After serving as resident physician in the Episcopal Hospital of Philadelphia, Burnett went abroad, spending ten months in post-graduate work. Returning to Philadelphia he practised for a year.
It was in Helmholtz's laboratory in 1871 that Burnett conducted his very valuable investigations into the condition of the membrane of the round window during the movements of the auditory ossicles and upon the results of changes in intralabyrinthine pressure. These were published in the Archives of Ophthalmology and Otology in 1872.
On his findings Burnett based the operation which he subsequently devised and performed in many cases for the relief of progressive deafness in chronic catarrhal otitis media, consisting in the performance of tympanotomy and removal of the incus. Burnett later advocated the same procedure for the relief of vertigo in chronic catarrhal otitis media.
In 1872 he entered into practise once more, now devoting his energies solely to diseases of the ear. In spite of the exacting claims of a very large practise, throughout his life he was able to devote much time to laboratory research and became known as one of the foremost investigators into the physiology of hearing. He was for many years professor of otology in the Philadelphia Polyclinic and served on the staff of several hospitals. Most of his operative work was done in the Presbyterian Hospital.
On January 15, 1902, he attended the meeting of the Section of Otology and Laryngology of the College of Physicians of Philadelphia and participated in the discussion of the papers read on that occasion. A few days later he developed pneumonia and died on January 30, at his home in Bryn Mawr.
Burnett, Charles Henry not only made very frequent contributions to periodical medical literature, but also edited the department of progress of otology in the American Journal of Medical Sciences. In 1877 he published The Ear: Its Anatomy, Physiology, and Diseases and in 1879 Hearing, and How to Keep It. In 1893 he edited a System of Diseases of the Ear, Nose and Throat. He also contributed the sections on otology in Keating's Cyclopedia of the Diseases of Children (1888 - 89) and in the American Textbook of Surgery (1892) and the American Year-book of Medicine and Surgery. In 1901 in conjunction with Dr. E. Fletcher Ingalls of Chicago, and Dr. James E. Newcomb of New York, he edited a Textbook of Diseases of the Ear, Nose and Throat. Burnett was a fellow of the College of Physicians of Philadelphia, a member of many other medical societies, and at one time president of the American Otological Society.

He was an indefatigable worker and must often have overtaxed himself with the combined labors of his practise and his scientific work.
He was a member of the American Otological Society and a member of many other medical societies.
Burnett was of frail physique and suffered much from ill health.
On June 18, 1874, he was married to Anna L. Davis.
