His research focused on the metabolism of carbohydrates and the structures of microbial cell walls. He joined the United States National Academy of Sciences in 1975. He served in the United States Navy from 1944 to 1946.
He held a postdoctoral fellowship with East. L. Hirst in the Department of Chemistry at the University of Edinburgh.
In 1955, Ballou was appointed to the biochemistry faculty at University of California, Berkeley, becoming Professor Emeritus in 1991. He served as an editorial board of the Journal of Biological Chemistry.
Served to Lieutenant junior grade United States Navy, 1944-1946. Guggenheim fellow, 1968.
