Denis Patrick O'Brien was born on March 24, 1939, in Knebworth, Hertford, United Kingdom. He is a son of Patrick Kevin O'Brien, an economist, and Dorothy Elizabeth (Crisp) O'Brien.
In 1960, Denis received a Bachelor of Science Degree in Economics from the University College London. Later, in 1969, he got his Doctor of Philosophy degree from Queen's University Belfast.
In 1963, Denis began his career as an assistant lecturer of Economics at Queen's University Belfast, a post he held till 1965, when he was appointed a lecturer in Economics. O'Brien held this post until 1970. During the period from 1970 till 1972, he served as a reader in Economics at the same university.
In 1972, O'Brien was appointed a Professor of Economics at Durham University. In 1997, he retired and was made a Professor Emeritus.

Denis married Eileen Patricia O’Brien on August 5, 1961. Their marriage produced three children — Ann Elizabeth Myerscough, Alison Mary Garnett and Martin Michael. Eileen Patricia O’Brien died in 1985.
Later, on September 11, 1993, Denis married Julia Stapleton, a university lecturer. Their marriage produced one child — Juliet Florence.
