Edward Delafield was born on May 7, 1794 in New York City, New York, United States. He was the third son among the eleven children of John Delafield, an English merchant who had emigrated to America, by his wife, Ann Hallett of New York.
He received his primary education first under a tutor named Adam Smith, and later at the Union Hall Academy under L. E. A. Eigenbrodt who gave him a good foundation in mathematics and the languages.
He graduated from Yale with the degree of A. B. in 1812, after which he became a pupil to Dr. Samuel Borrowe of New York, under whose tutelage he obtained, in 1816, the degree of Doctor of Medicine from the College of Physicians and Surgeons, New York, for which he wrote An Inaugural Dissertation on Pulmonary Consumption (1816).
Delafield went to Europe with John Kearny Rodgers where he had a year (1817) in the clinics of England, Scotland, Holland, and France.
On returning to New York, though he continued in general surgery and obstetrics, his attention was directed chiefly to the diseases of the eye.
He edited in 1825 an American edition (based on the third London edition) of Benjamin Travers’s Synopsis of the Diseases of the Eye, and Their Treatment, with notes and additions, and he later made important contributions to ophthalmological journals.
In 1818 he formulated a plan for establishing the New York Eye Infirmary modeled after the London Infirmary in which he had studied, and succeeded in carrying it out in association with Kearny Rodgers in 1820; two rooms were opened, and during the first seven months 436 patients were treated. Later, largely through Delafield’s influence, more ample accommodations were secured. Between 1820 and 1906 over a million patients were treated.
In 1824 the Massachusetts Charitable Eye and Ear Infirmary was founded in Boston by Edward Reynolds and John Jeffries, who had received their stimulus from New York.
He was also largely responsible for founding a society in 1842 for the relief of the widows and orphans of medical men, which also was based upon a similar society then existing in London.
In addition to his labors at the Eye and Ear Infirmary, he occupied from 1825 until 1838 the chair of obstetrics and diseases of women and children at the College of Physicians and Surgeons of New York, and from 1858 to 1875 he was president of the College.
As a practitioner he was unusually successful and is said to have possessed to a remarkable degree the power of inspiring the confidence of his patients.

Delafield was a genial and kindly man, and owing to his long gray beard and sideburns was rather striking in personal appearance.
In October 1821 Delafield married Elina Langdon Elwyn, grand-daughter of John Langdon, governor of New Hampshire and president pro tempore of the Senate in the First Congress of the United States. There were six children, all of whom died before their father. In January 1839 he married his second wife, Julia Floyd, grand-daughter of William Floyd, signer of the Declaration of Independence; she became the mother of Francis Delafield, the pathologist.
