She was a daughter of Landgrave Henry III of Upper Hesse (nicknamed Henry the Rich) and his wife, Anna of Katzenelnbogen.

On 11 February 1481, Elisabeth married Count John V of Nassau-Dillenburg. Together, they had six children:

Elisabeth died in Cologne in 1523 and was buried in Siegen.
