Emi Sampaku was born in Aki Province (Hiroshima Prefecture), he was bereaved of his parents when young and brought up by a family called Fujioka. His family name was Tsutsumi.

He studied hard the classical medical book, Shokanron, written by a Chinese, Chang Chung-ching.
Sampaku was an advocate of classical Chinese medicine. In his time, Todo Yoshimasu came out with a theory that all diseases are caused by one poison and Sampaku supported it. Sampaku became famous as a physician and was employed by the Lord of Aki, receiving a fief of 200 koku of rice.
He became the son-in-law of Ryogcn Emi.
