Klebe was born on April 19, 1954 in Frankfurt, Germany; the son of Dieter and Lieselotte (Becker) Klebe.
Klebe received a diploma in chemistry from the University of Frankfurt in 1977. Five years later he earned his Ph.D. from the University of Frankfurt.
Klebe began his career as a teacher of physical chemistry at the University of Frankfurt in 1978 and held the position for two years. In 1981, he dealt with neutron scattering and differential electron determination in crystallography at the Institut Laue-Langevin. Then in 1982, Gerhard took a position of a researcher at the Institute of Crystallography of the University of Frankfurt. A year later he became a postdoctoral researcher at Chemical Crystallography Laboratory of the University of Bern.
In 1984, Klebe was appointed a research scientist in the field of drug design and crystallography at the main laboratory of BASF SE, where he worked until 1996. Also he served as a lecturer at the University of Heidelberg from 1992 to 1996. Since 1996 Gerhard has been a professor of pharmaceutical chemistry and drug design at Philipps University of Marburg.
In addition, he was an editor of the Journal of Computer-Aided Molecular Design. Klebe is on the board of the Cambridge Crystallographic Data Center.

2013On April 10, 1989 Gerhard Klebe married Gabriele Metzler. They have a child.
