Seki was born on December 14, 1937 in Okayama City, Japan; the son of Masaji and Sakae (Sentou) Seki.

Seki received a Bachelor's degree in agriculture in 1961 and a Master's degree in agriculture in 1963 from the University of Tokyo. Also in 1966, he was given a Doctor of Philosophy degree in agriculture from the same university.
Seki began his career as a research associate at the University of Tokyo in 1966 and held it for ten years. In 1973, he took a position of a lecturer at Waseda University, where he worked until 1975. A year later Humitake was appointed an associate professor of microbiology at the University of Tsukuba. Then in 1989, he became a professor at the same university and held it for twelve years.
In addition, Seki worked as a dean of master's program in biosystem studies at the University of Tsukuba from 1998 to 2001. Since 2001 he has been a professor emeritus at that university. In 2004, he served as a lecturer at Miyazaki University. A year later Humitake took the same position at Ocean University of China and at Yamagata University in 2008.
Seki is a member of Japanese Academy of Budo, American Society of Microbiology, Oceanographic Society of Japan and Japan Science Council.
On May 31, 1970 Humitake Seki married Atsuko Yamamoto. They have 2 children.
