She is best known for her lead role on Homicide: Life on the Street as Megan Russert and supporting roles on Judge Advocate General as Meredith Cavanaugh, Dear John as Kate McCarron, and on Beggars and Choosers as Cecile Malone. Hofmann performed with the comedy troupe The Second City before breaking into television in 1986. She attended Columbia College Chicago.

She has appeared twice in Season 7 of television series Criminal Minds as David Rossi"s first ex-wife Carolyn Baker.
