Burbridge was born on September 22, 1961 to William and Carol Burbridge in the Bronx, New York, although the family moved to Washington, District of Columbia two years later.

Burbridge is a classically trained multi-instrumentalist, and he has provided keyboards, organ, flute, and backing vocals for various bands throughout his career. Kofi is also the brother of bass player Oteil Burbridge, who is known for his work in the Allman Brothers Band and Aquarium Rescue Unit, and who also used to be part of the Tedeschi Trucks Band before leaving the band on October 5, 2012. The band broke up after less than a year but introduced him to other notable musicians in the area like Bruce Hampton.
He eventually joined the Aquarium Rescue Unit shortly after Colonel Bruce Hampton left, which reunited him with Oteil Burbridge and Jeff Sipe.
The Derek Trucks Band
Burbridge joined the band in 1999. In an interview upon the release of their 2002 album, Joyful Noise, Trucks commented in 2006, "Kofi Burbridge has been with us maybe 2–3 years, and he"s one of the few musical geniuses that I"ve had the chance to work with.." continuing, "I"m really anxious to see in the next few years to see where he takes this, because he"s definitely a huge part of what"s going on right now."
The Tedeschi Trucks Band
Tedeschi Trucks Band performed at Eric Clapton"s Crossroads Guitar Festival 2010 and appears on the Digital Video Disc release with two of their songs, "Midnight in Harlem" and "Comin" Home".
In 2012, the band released their second album, Everybody"s Talkin", a double live album compiled from their 2011 world tour. On August 2013 their third album Made Up Mind, their second studio album, was released.
