Lorraine Dennerstein received an Bachelor of Medicine and Bachelor of Surgery from the University of Melbourne in 1970. She was awarded a Doctor of Philosophy from the department of Obstetrics and Gynaecology at Monash University in 1979 for her study of the effects of the contraceptive pill on libido in women. Subsequently, she qualified as a psychiatrist in 1980, obtained a Diploma in Psychological Medicine from the University of Melbourne in 1981 and became a fellow of the Royal Australian and New Zealand College of Psychiatrists in 1983.
