Untermeyer was born on October 1, 1885 in New York City, the son of a jewelry manufacturer.
Untermeyer was, for the most part, self-educated.
For more than 20 years Untermeyer was engaged in the jewelry manufacturing business, devoting his evenings to poetry. After 1923, when he retired from business, he devoted himself to writing, editorial work, and lecturing. Best known as a maker of anthologies, he edited many collections of verse, including This Singing World (1923), Modern American Poetry (1962), and Modern British Poetry (1962). His own poetry was collected in Roast Leviathan (1923) and Burning Bush (1928). Other works include Bygones: The Recollections of Louis Untermeyer (1965) and The Pursuit of Poetry (1969). Untermeyer died in Newtown, Connecticut, December 18, 1977.

Untermeyer married Jean Starr in January 1907, and their son Richard was born in December of that year. After a 1926 divorce, they were reunited in 1929, after which they adopted two sons, Laurence and Joseph. He married the poet Virginia Moore (1903-1993) in 1927; their son, John Moore Untermeyer (1928), was renamed John Fitzallen Moore after a painful 1929 divorce. In the 1930s, he divorced Jean Starr Untermeyer and married Esther Antin (1894-1983). This relationship also ended in divorce in 1945. In 1948, he married Bryna Ivens, an editor of Seventeen magazine.
