Khalifa was born on January 27, 1964 in Cairo, Egypt; the son of Ibrahim Mohamed Khalifa and Tahany Mostafa Fahmy.

Khalifa received a Bachelor's degree in Pharmaceutical Sciences from King Saud University in 1986. Eight years later he was given a Doctor of Philosophy degree from the University of Mississippi.
Khalifa began his career as an associate professor at Suez Canal University in 1995 and held it for eleven years. In 2001, he was appointed an adjunct associate professor at the University of Mississippi, where he worked until 2005. Also in 2002, Sherief was a visiting professor at the University of Georgia.
Then in 2004, Khalifa held the position of a medical consultant at Inter Mark Advertising & Consultancy. In addition, he became an acting head of the department of pharmacy practice and clinical pharmacy at Misr International University in 2007. The same year he served as a director of medical education department at Medex-Middle East. Since 2010 Sherief has been a professor and associate dean at College of Pharmacy of Gulf Medical University.
Khalifa is a member of Egyptian Pharmaceutical Society and Egyptian Syndicate of Pharmacists.
On February 13, 1972 Sherief Khalifa married Samah Abdel-Rassoul El-Okda. They have 3 children.
