Thyra Holt worked at the Munich photographic studio of Wanda Debschitz, who had a large influence on Holt's development as a photographer. She also served as apprentice for a year with Francis de Jongh in Lausanne, Switzerland.
Thyra Holt established herself as a photographer in Kolding, Denmark, in 1921, and from 1928 to 1958 operated a studio at Amagerterv, Copenhagen, with Margrethe Madsen. She and her partner photographed a number of authors, such as Pearl S. Buck and Rabindranath Tagore.
