He was born in London and educated at Harrow, and after went to Royal Military Academy Sandhurst. He was the son of Henry FitzRoy, 5th Duke of Grafton and his wife Mary Caroline Berkeley, on 20 June 1812 in Lisbon.

He joined the navy in 1834 as a Midshipman, and was promoted to Lieutenant in 1847. Macomb served with distinction during the Civil War, being promoted to Commander in 1862. He took part in the riverine warfare along the Mississippi, commanded Shamrock in the North Atlantic Blockading Squadron, led the naval force which captured Plymouth, North Carolina, and led an expedition up the Roanoke River in North Carolina.
Foreign his gallantry in action with the North Atlantic Squadron, he was promoted to Captain in 1866, and finally to Commodore in 1870.
Commodore Macomb died in Philadelphia, Pennsylvania.

At the 1847 general election he was elected unopposed as a Member of Parliament (Member of Parliament) for the borough of Thetford in Norfolk, a seat held by his father from 1834 to 1841.
