Knollys was the oldest 1st surviving son of Charles Knollys and his wife Elizabeth, daughter of Michael Lister of Burwell, Lincolnshire.

However, the father"s claim had been rejected by the House of Lords in 1697. He held the seat until his death in June 1740, pre-deceasing his father.
