Whitmore was the son of William Whitmore merchant of London and his wife Anne Bond, daughter of alderman William Bond.

He inherited the estate of Apley, Shropshire on the death of his father in 1593. He was Sheriff of Shropshire in 1620. He was knighted at Greenwich on 24 or 28 June 1621.
In 1624 he was re-elected Member of Parliament for Bridgnorth.
He was elected Member of Parliament for Bridgnorth again in 1626. Whitmore died at the age of 76.
He had three sons of whom the eldest Thomas was Member of Parliament and was created a baronet.
