Shelepin graduated from the Moscow institute of Philosophy, Literature and History named after N. G. Chernyshevsky in 1941.
In 1939 Shelepin joined the Red Army. Participant of the Winter War of 1939-1940, deputy political instructor, political commissar of squadron. Since 1940 he served as instructor, head of department, secretary of the Moscow City Committee, All-Union Leninist Young Communist League. Since 1943 he was a secretary, 1st secretary of the Central Committee, All-Union Leninist Young Communist League. Since 1958 - the head of the Party Organs department of the Central Committee of CPSU. Since 1958 the chairman of the Committee for State Security under the Council of Ministers of USSR. Since 1961 - secretary of the Central Committee of CPSU, at the same time in 1962 – 1965 the chairman of the Committee of Party and State Control under the Central Committee of CPSU and the Council of Ministers, vice-chairman of the Council of Ministers of USSR. Since 1967 - the chairman of the All-Union Central Council of Trade Unions. In 1975 – 1984 he was the vice-chairman of USSR State Committee on Technical and Vocational Education. In 1964 – 1975 Shelepin was a member of the Presidium (Political Bureau) of the Central Committee of CPSU. Deputy of the Supreme Soviet of the USSR (1954-1978).
