Cole served in the United States Army Air Force from 1942 to 1946. In 1953, he began his career as an instructor of physiology at Duke University. A year later Benjamin took a position of an assistant professor at Louisiana State University. Then in 1959, he became a researcher of biology division at Oak Ridge National Laboratory. Also Cole served as an associate professor of biology at the University of South California from 1960 to 1963.
In 1963, he was appointed a professor at the University of South California. From 1987 Benjamin was a distinguished professor emeritus at the same university.
Cole was a member of the American Association for the Advancement of Science, American Physiological Society, Society of Experimental Biology, Medical Association of Southeastern Biologists and Sigma Xi.
On May 30, 1943 Benjamin Cole married Leona Todd. They have two children.
