Johnson was born on September 23, 1873 in Concord, Indiana, United States; the son of Frederick Angell Johnson and Marie Jane (Tustison) Johnson.
Johnson received a Bachelor of Science degree in 1900 and a Doctor of Philosophy degree in 1903 from the University of Michigan.
Johnson began his career as an assistant instructor of chemistry at the University of Michigan in 1898. Three years later he took a position of an instructor of chemistry at the University of Iowa and held it for a year.
Also Charles served as an assistant professor of chemistry at the University of Washington from 1903 to 1904. Then in 1904, he became a professor of pharmaceutical chemistry and dean of Pharmacy Department at the same university. Since 1939 Johnson held the position of a professor and dean emeritus at the University of Washington.
Johnson was a member of American Pharmaceutical Association (president, 1927-1928), Washington State Pharmaceutical Association, Kappa Psi, Sigma Xi, Rho Chi and Phi Delta Chi.
On June 21, 1900 Charles Johnson married Parthenia Sykes. Then on June 22, 1920 he married Frances Edith Hindman. They have 3 children.
