Freifelder was born on July 19, 1935 in Philadelphia, Pennsylvania, United States; the son of Morris Leon Freifelder and Florence (Levenson) Freifelder.
Freifelder received a Bachelor of Science degree in Physics from the University of Chicago in 1957. Two years later he earned his Doctor of Philosophy degree in Biophysics from the same university.
Freifelder began his career as a research associate at Massachusetts Institute of Technology in 1960. Two years later he took the same position at the University of Copenhagen. In 1963, David became an assistant professor of medical physics at the University of California, Berkeley.
He was a member of faculty at Brandeis University from 1966 to 1983. In addition, in 1976, Freifelder was appointed a professor of biochemistry at the same university, where he worked until 1982. He was also a visiting scholar at the Weizman Institute and Stanford University.
