John studied law with James M. Bethune.
John Lamkin was admitted to the bar in 1833. On Christmas Day 1835, the Lamkins moved to Texas, and they later went to New Orleans, where Lamkin became an accountant.
In 1838, he began a law practice in Pike County, Mississippi. From 1841 to 1845, he was a district attorney in Holmesville, Pike County. He also owned a modest farm.
Lamkin seems to have spent the last years before the war in pursuit of his law practice. In April 1862, he raised the Holmesville Guards of the 33rd Mississippi Regiment, serving as a captain.
On October 5, 1863, he was elected to the second Confederate House, defeating the radical secessionist John J. McRae for Congress. He served on the Commerce, Patents, and Post Office and Post Roads Committees.
Lamkin nevertheless acquiesced in support of the Confederacy after his long opposition to secession had failed. He opposed the Davis administration, though he did not join the union faction in Congress.
On November 14, 1835, John married Thurza Ann Kilgore. They had five children.
