John attended local public schools and taught public school before studying law at Winchester Law School from 1828 to 1829.
A lawyer who also planted, Chambliss resided in Hicksford, Greensville County, Virginia. He was elected commonwealth attorney for the county in 1849.
He opposed secession at the Virginia secession convention. Chambliss was elected to the first Confederate Congress, where he served on the Committee on Naval Affairs from 1862 until 1864.
Chambliss also served on the special committee on the veterans' home for wounded soldiers of the Army of Virginia. He refused to run for reelection.
A man with little political experience, Chambliss spent the rest of his life paying off debts incurred during the war and again practiced law.
John was a member of the Baptist Church.
Chambliss was a vigorous supporter of the Davis administration and its policies on building gunboats for the protection of the Virginia coastline.
John married Tilly Woodruff and they had seven children. His son, John Randolph Chambliss, Jr., was a colonel in the 13th Virginia Cavalry Regiment.
