Kunio Tsuji was born on September 24, 1925 in Tokyo, Japan. He was a son of Seigo Tsuji, a journalist and musician, and Kimi (Yuda) Tsuji, a doll maker.

Kunio Tsuji attended Matsumoto Agatagaoka High School with Kita Morio. He studied French literature at the University of Tokyo, received his Master of Arts in 1952 and finished his postgraduate studies in 1952-1954. Besides, Kunio Tsuji attended the Sorbonne, Paris in 1957-1961.
After graduation, Kunio Tsuji became an instructor at Gakushūin University and a literary critic. He spent the years 1957-1960 in France, which strongly influenced his development as a novelist.
In 1963 he published his first mature work "Kairō nite" ("In the Corridor").
Some of his more celebrated later novels include "Azuchi ōkanki" (translated as "The Signore") and "Haikyōsha Yurianusu" ("The Apostate Julianus").
Tsuji's works were on the whole idealistic and spiritual. They included many historical novels in which the protagonists search for the meaning of life at times of great social change.
Kunio Tsuji was an assistant professor of literature at St. Paul’s University (now Rikkyo University) in Tokyo during 1966-1972. He then worked as a professor of literature at Tokyo Agricultural College in 1972-1975. From 1975 to 1990 Kunio Tsuji was a professor of French literature at Gakushūin University.
Besides, he was a lecturer in cultural anthropology at the University of Paris X in 1980-1981 and a lecturer in Japanese literature at the University of Paris III from 1983 to 1984.
Kunio Tsuji was a member of Japan Writers Association, Academy of Japan.
Kunio Tsuji married Sahoko Goto, an art historian, on June 6, 1953.
