Lagorio was born in Feodosiya on December 9, 1826. He was the son of Felice Lagorio, who was a Genoese merchant serving as Vice-Consul for the Kingdom of the Two Sicilies.

Lev Lagorio took private lessons with Ivan Aivazovsky from 1839 to 1840. With the support of Alexander Kaznacheyev, the Governor of Taurida, he was able to enroll at the Imperial Academy of Arts (today known as Ilya Repin St. Petersburg State Academic Institute for Painting, Sculpture and Architecture) in 1842. Later, he received financial assistance from the Duke of Leuchtenberg, the Academy's new President. He was taught there by Alexander Sauerweid, Bogdan Willewalde and Maxim Vorobiev.
In 1850, Lagorio obtained the title of "Artist" for his painting "View of Lahti". Two years later, he became a Russian citizen. Moreover, he was also awarded a pension to study abroad. First he visited Paris in 1853, where he studied paintings by French masters in the Louvre and the Luxembourg Palace. Then he went to Rome, staying there until 1859, after having received permission to extend the stay abroad at his own expense for a period of two years.
In 1860 Lagorio came to St. Petersburg. The same year he became a professor of landscape painting and also exhibited the works he had created in Italy. In 1861 Lev Lagorio made a trip to the Caucasus and presented his series of landscapes from there to Tsar Alexander II.
In 1863 Lev Lagorio accompanied the Grand Duke Mikhail Nikolayevich on his trip to the Caucasus. There he participated in the Caucasian War. After the war, he settled in Saint Petersburg and spent summer months in Sudak, where he opened a studio. In 1885 he visited Constantinople.
In the year of 1885 he was commissioned by Alexander III to create a series of paintings devoted to the Russo-Turkish War of 1877-1878. To make it he visited battlefields throughout the European and Asian theaters.
Lev Lagorio was one of the greatest painters of the second half of the XIX century in Russia. His works are in many museum collections, including the State Tretyakov Gallery, State Russian Museum, the Scientific Research Museum of Russia Academy of Arts, GMII im. Pushkin and others.
In 1861 Lev Lagorio was presented with the Order of Saint Anna by Tsar Alexander II. In 1890 he became one of the founders of the St. Petersburg Society of Artists. In 1906 St. Petersburg held a memorial exhibition of the master.
