Tozzi was born in Fossombrone, Italy, on October 30, 1895. His father, Giacinto Tommaso, was a doctor. Mario Tozzi was the eldest of five brothers.

Initially, Mario Tozzi began to study chemistry before entering the Accademia di Belle Arti di Bologna in 1913. There he got acquainted with Giorgio Morandi and Osvaldo Licini. He graduated from the academy in 1915.
Tozzi served during the First World War, in which he lost two of his brothers. In 1919 he settled with his wife in Paris. There he exhibited at the Salon des Artistes Indépendantes, at the Salon d'Automne and at the Salon des Tuileries where he was immediately noticed by the art critics. In 1926 he met a lot of avant-garde Italian painters who had a strong influence of him, in particular, with Severo Pozzati. The same year, he exhibited at the first Novecento exhibition. He founded the Groupe des Sept (also known as Les Italiens de Paris) with Massimo Campigli, Giorgio de Chirico, Filippo de Pisis, Renato Paresce, Alberto Savinio, and Gino Severini.
He moved back to Rome in 1936 and devoted himself to producing frescoes. Mario Tozzi exhibited at the Venice Biennale in 1938 and 1942. During the forties and fifties, he almost didn't work, because of serious health problems. However, he participated at the Venice Biennale in 1948, 1952 and 1954.
Eventually, Tozzi presented his works at the Annunciata gallery in Milan in 1958. In 1960 he settled in Suna in the family home. He returned to France in 1971 to be close to his daughter and grandchildren, and lived there until his death.
