Mikisaburo Rai was born in Osaka, Japan in 1825. He was the third son of Sanyo.
When a youth, he lived in Osaka and studied at the Shoheiko (Shogunate college) in Edo (now Tokyo). He was a friend of Issai Sato and Gozan Kikuchi.
Mikisaburo Rai traveled widely in Japan, going as far as Hokkaido and Kyushu. He settled in Kyoto and taught Chinese literature to youths. He worked for the cause of reform of Shogunate administration and expulsion of foreigners together with Seigan Yanagawa and Umpin Umeda. Involved in the Ansei-no-Taigoku (Ansei Era Trouble), Mikisaburo Rai was arrested and sent to Edo where he was beheaded.
