Myddelton was the son of Sir Thomas Myddelton, 1st Baronet of Chirk Castle and his first wife Mary Cholmondley, daughter of Thomas Cholmondley of Vale Royal, Cheshire.

He succeeded to the baronetcy of Chirke in the County of Denbigh on the death of his father in 1664. In 1679, Myddelton was elected Member of Parliament for Denbighshire and sat until 1681. Myddelton married firstly Elizabeth Wilbraham, daughter of Sir Thomas Wilbraham and Elizabeth of Weston Park,Weston-under-Lizzard,Staffordshire.(She Died in childberth aged 22 in 1675He sent to Weston for her portrait so that the sculptor, Bushell,in Chester could make a monument to herThe monument is in Chirk churchyard,the portraite never came back to Weston and is still missing!)and secondly Charlotte Bridgeman, daughter of Sir Orlando Bridgeman, 1st Baronet.
