Sadatsugu Tsutsui was born on June 6, 1562 in Jimyoji Castle at Yamato (Nara Prefecture). He was the son of Yorikuni Jimyōji. Around 1571 he was adopted by his uncle Junkei, a feudal lord of the Yamato province.
After the death of Junkei in 1584, Sadatsugu succeeded to his father-in-law's estate. The following year he was transferred by Hideyoshi Toyotomi to Ueno Castle (Mie Prefecture) and conferred a fief of 200,000 koku, but during the Sekigahara Battle (1600) he distinguished himself on the side of Ieyasu Tokugawa. Because of his friendship with Harunaga Ōno, a general of Toyotomi, Sadatsugu Tsutsui was suspected by Tokugawa and confined to his castle. Later his property was confiscated and he was exiled to Taira (Fukushima Prefecture) (1608).
Sadatsugu Tsutsui was married to one of Oda Nobunaga's daughters, Hideko.
