He was the youngest son of Gustavus Hamilton, 1st Viscount Boyne and his wife Elizabeth Brooke, daughter of Sir Henry Brooke.

In 1725, he entered the Irish House of Commons for Street Johnstown (County Donegal), a seat he held until 1727. He was Collector of the Portuguese of Cork.
Three years later, he was returned for Donegal County, the same constituency his father and his two older brothers Frederick and Gustavus had also represented, and sat as Member of Parliament until his death in 1743.
