Bost was born on January 5, 1901 in Rockwell, North Carolina, United States; the son of James Walton Bost and Mary Lee (Miller) Bost.
Bost received a Bachelor of Arts degree from Newberry College in 1923. A year later he earned his Master of Arts degree and a Doctor of Philosophy degree in 1928 from the University of North Carolina.
Bost began his career as an assistant in chemistry at the University of North Carolina in 1923. A year later he took a position of an instructor of chemistry at Tulane University.
He worked as an instructor of chemistry at the University of North Carolina from 1926 to 1928. Then in 1928, Ralph became an assistant professor and an associate professor in 1934 at the same university. Since 1937 he was a professor at the University of North Carolina. Also Bost served as a head of chemistry department at that university since 1939.
Bost was a member of American Chemical Society, Chemical Society of London, Elisha Mitchell Scientific Society, American Physical Society, North Carolina Academy of Science, Pi Kappa Delta, Sigma Xi, Kappa Psi and Alpha Chi Sigma.
On July 12, 1927 Ralph Bost married Beulah Christine Cauble.
