Mendis is the daughter of Marthelis and Mary Winifred Baduge.
Mendis received a Doctor of Medicine degree from the University of Colombo in 1974.
Mendis began her career as a senior house officer in Bristol in 1975 and had held the position for four years. In 1988, she became a research fellow at the University of Bristol, where she worked until 1989. In 1994, Shanthi became a professor of medicine at the University of Peradeniya and had held the position for six years.
In addition, she served the World Health Organization as a senior adviser, cardiovascular disease and other noncommunicable diseases for 20 years. During her tenure in that organization Shanthi has led and overseen the development of global action plans in 2013, global reports in 2014, global conferences and many technical guidelines on noncommunicable diseases. Mendis also was responsible for the implementation of innovative multi-country projects on noncommunicable diseases to strengthen primary health care policy and research capacity in developing countries.
Nowadays she is an independent consultant in Global Health and adviser at the Geneva Learning Foundation.

2017Mendis is a member of the Royal College of Physicians of London and Edinburgh and the American College of Cardiology. In 2014, she was elected to life-membership of the World Stroke Association.
Shanthi Mendis is married to Ranjit Rigobert Mendis. They have two children.
