He was the son of Sir John Barker, 4th Baronet and Bridget Bacon, daughter of Sir Nicholas Bacon.

He was educated at Pembroke College, Cambridge.
In 1696, he succeeded his father as baronet. Barker entered the British House of Commons for Ipswich in 1708, representing it to 1713. Barker married firstly Mary Bence, only daughter of John Bence, and by her he had an only child.

He was subsequently Member of Parliament (Member of Parliament) for Thetford until 1715 and for Suffolk from 1722 until his death in 1731.
