Biles was born on November 20, 1950 in Urbana, Illinois, United States; the son of Lloyd Wilson Biles and Irene Frances (Brawner) Biles.

Biles received a Bachelor of Arts degree from the University of Illinois in 1972. Two years later he earned his Master of Arts degree and a Doctor of Philosophy degree in 1981 from the same university.
Biles began his career as an instructor at Parkland College in 1975 and held the position for two years. In 1981, he took a position of a visiting assistant professor at Memphis State University (nowadays University of Memphis). Three years later William was appointed an assistant professor at Oklahoma State University. From 1986 he was an associate professor at the same university. Nowadays Biles has been a professor at Oklahoma State University.
Biles is a member of Organization of American Historians, Southern History Association, Urban History Association, Phi Beta Kappa, Phi Kappa Phi and Phi Eta Sigma.
On December 17, 1977 William Biles married Mary Claire Frank. They have three children.
