Yoshiharu Ashikaga was born in 1511. He was the son of the eleventh shōgun Ashikaga Yoshizumi. His childhood name was Kameomaru.

After Shogun Yoshitane's downfall, he was proclaimed Shogun with the support of Takakuni Hosokawa (1521). During his rule, the Miyoshi Family and its followers and bandits gripped Kyoto, disdaining the Shogunate orders. As Shogun, Yoshiharu was forced to flee from Kyoto three times. He turned over the Shogunate to his son, Yoshiteru, in 1535.
