Quack was born on July 22, 1948 in Darmstadt, Germany.
Quack studied at Technische Universität Darmstadt from 1966 to 1969. Also he studied at the University of Grenoble (nowadays Université Grenoble Alpes) in 1969-1970. In 1972, Martin entered the Swiss Federal Institute of Technology, where he obtained a doctorate in 1975.
Quack began his career as a Max Kade Fellow at the University of California, Berkeley in 1976 and had held the position for a year. In 1978, he was appointed a privatdozent at the University of Gottingen. Then in 1982, Martin became a professor at the University of Bonn.
Since 1983 he has been a professor of physical chemistry at ETH Zurich. In addition, Quack was Head of the Laboratory of Physical Chemistry at that university in 1986-1987, 1991-1992 and again from 2006 to 2007. In 2005, he took the position of Miller Visiting Research Professor at the University of California, Berkeley.

Quack is a member of the American Physical Society, the Academy of Sciences Leopoldina, the Berlin-Brandenburg Academy of Sciences and Humanities, the American Academy of Arts and Sciences and Göttingen Academy of Sciences and Humanities.
He was a member of the National Research Council of the Swiss National Science Foundation from 2002 to 2011. In 2014, Martin was elected as a member of the presidium of the German Academy of Sciences, Leopoldina.
On May 24, 1977 Martin Quack married Roswitha Quack. They have three children.
