Sen was born in Dhaka, Bangladesh, on October 18, 1918. He was one of 20 children in a family of ayurvedic healers.
Paritosh Sen had a Diploma in Fine Arts from the Government College of Fine Arts, Chennai. In 1949 he went to Paris to study further, receiving his artistic training at the Academie Andre Lhote, the Academie la Grande Chaumiere, the École nationale supérieure des Beaux-Arts, and the École du Louvre in Paris.
Sen moved from Paris to Calcutta in 1942, where he and a group of friends formed the Calcutta Group. It was an association of artists that sought to incorporate contemporary values in Indian art. In Calcutta, he taught first at Babasaheb Bhimrao Ambedkar Bihar University and then at Jadavpur University. He also worked at The Daly College at Indore during the late 1940s.
While in Paris, Paritosh Sen met Pablo Picasso and Constantin Brâncuși and fell under the influence of both Cubism and Expressionism, though he never moved into fully abstract work. He moved back to India in 1954 and drew his inspiration from the life of Calcutta.
A prolific writer, Paritosh Sen has published many works in both Bengali and in English, including a series of autobiographical vignettes titled "Jindabahar Lane". In 1959-1960, he published "Zindabahar", a book of autobiographical sketches in which he memorialized the Dhaka city of his childhood.
In 1981-1982 he was the artist-in-residence at the Maryland Institute College of Art. Four years later he accepted a similar position at the National Institute of Design at Ahmedabad, in Gujarat, where he taught a course in illustration.
