In 1990 he graduated from the secondary school named after V.M. Komarov in the town Zvezdniy and entered the Tambov Higher Military Aviation School for pilots named after M.M. Raskovoy and graduated in 1995 with the specialty "Command tactical aviation" and the assignment of qualification "pilot-engineer".
On March 5, 1996 he became a pilot and since April 30, 1996 he was working as an assistant commander of the ship of the air squadron of control and retransmission 353 special aviation regiment 8 special-purpose air mission (Chkalov division).
On July 28, 1997, by decision of the GMPC was recommended for the position in the cosmonaut corps of the Air Force CTC (12th set).
From December 1997 to November 1999 passed the course of general space training.

From January 2005 to February 2006 he was a part of cosmonauts group tested for flights to the ISS. In the period from February to April 2006 he was trained as a backup engineer of ISS-13 / EP-10 as a flight engineer-2 of Soyuz TMA and the flight engineer for the visiting expedition to the ISS. From May 2006 to April 2008, he was preparing for a space flight as part of the ISS-17 main crew as commander of the Soyuz TMA and the ISS commander. Since December 2008 – Sergey Volkov had been the instructor-test-cosmonaut of the 2nd class of cosmonaut detachment of "Ya.A.Gagarin Research&Test Cosmonaut Training Center".From December 2009 to October 2010, was trained as a backup crew of ISS-25/26 as commander of Soyuz TMA-M and ISS flight engineer.
Since September 2010 - the instructor-cosmonaut-test-deputy commander of the cosmonaut detachment of the Federal State Institution of the Russian Federation " Ya.A.Gagarin Research&Test Cosmonaut Training Center".
From October 2010 to June 2011, he was trained in the ISS-28/29 main crew as the commander of the Soyuz TMA-M and the ISS flight engineer.
From August 2012 to April 2014 he worked as the instructor-cosmonaut-tester-commander of the detachment of cosmonauts Roscosmos.
In October 2013 he became the instructor-cosmonaut-tester of the 1st class in Roskosmos. From November 2012 to March 2015 he was trained as a backup crew of ISS-43/44 as a flight engineer of the Soyuz TMA-M and ISS flight engineer.
From October 2014 to September 2015 he was trained as a commander of the Soyuz TMA-M and ISS-45/46 flight engineer in the ISS-45/46 / EP-18 main crew.
From April 8 to October 24, 2008 Volkov fulfilled his 1st space flight as commander of the Soyuz TMA-12 spacecraft and the expedition commander of the ISS-17 together with Roskosmos cosmonaut O.D. Kononenko and Li So-yon - participant in the space flight. The installation of the main sealed section JEM PM of the research module "Kibo" and the manipulator JEMRMS was carried during the flight and the work was performed with the first cargo ship ESA ATV-001 "Jules Verne".
