Takatsugu was born in 1601 in Japan. He was the eldest son of Takatora, the Lord of Tsu Clan (Mie Prefecture).

Takatsugu Todo succeeded to the family estate upon the death of his father (1630). When Shogun Iemitsu Tokugawa traveled from Edo to Kyoto he accompanied him as a bodyguard (1634).
Later was appointed Chamberlain, and then placed in charge of the construction work of a stonewall of Edo Castle and also of the mausoleum at Toeizan, Edo. Subsequently was promoted major general.
