William R. Elton was born on August 15, 1921, in New York. He was the son of William and Mollie Elton.
In 1941, Elton received a Bachelor of Arts from Brooklyn College. In 1942, he received a Master of Arts from the University of Cincinnati, and then, a Ph.D. from the Ohio State University in 1957.
From 1945 to 1946, Elton was an instructor in English at Ohio State University in Columbus, and then, at Brown University in Providence, Rhode Island, he was an instructor in English from 1946 to 1950. From 1950 to 1951, he was an instructor in English in New York University and then, a visiting assistant professor of English at University of Connecticut in Storrs from 1952 to 1953. He also was an assistant professor of English at Ohio State University from 1953 to 1955.
From 1955 to 1969, at the University of California in Riverside, Elton began as an assistant professor, and then became a professor of English. In 1969, he became a professor of English at City University of New York. In 1951, at Oxford University he was a lecturer at Lincoln College, and then he was a Fulbright lecturer in India, in 1961. In 1969, he was a visiting professor at Columbia University, Institute for Advanced Study in Princeton, New Jersey, and first visiting Mellon Professor from 1984 to 1985. At the University of Paris VII Elton was a lecturer at Ecole Normale Supérieure in 1990, and lecturer at New College in 1990.

Elton was a member of the International Society for Neoplatonic Studies, International Association of Professors of English, Modern Language Association of America and Renaissance Society of America.
He also was a member of the Malone Society, Shakespeare Association of America, American Philological Association, Society for Ancient Greek Philosophy, Society for Renaissance Studies (England), PEN Club and English Institute as well.
In 1970, Elton married Mary Elizabeth Bowen, but they divorced in 1974.
