Kestler received a Bachelor of Science degree from Gonzaga University in 1970. Four years later he earned his Doctor of Philosophy degree from the University of Arizona.
Kestler began his career as a postdoctoral research associate in the department of botany at the University of Georgia in 1975. A year year later he took a position of a postdoctoral research fellow in the department of oral biology and orthodontics at the University of Connecticut Health Center. Then in 1979, Daniel was appointed an assistant research scientist in the department of cell and molecular biology at Medical College of Georgia.
In 1987, he held the same position at Guthrie Research Foundation. A year later Kestler became a research associate at Indiana University School of Medicine. He worked as a research scientist at Donald Guthrie Foundation for Medical Research from 1990 to 1992.
In 1992, Daniel was a research associate at the University of Tennessee Graduate School of Medicine. Since 1999 he has been an assistant professor of medicine at the same school.
On July 5, 1980 Daniel Kestler married Beth Kestler, with whom he divorced in 1990. They have a child. On July 12, 1991 he married Donna Maria Kestler.
