Use of nanotechnological based formulations and nanomaterials are increasing day-by-day in wide range covering a broad typology of applications, from design and development of targeted drug delivery systems, manufacturing of pesticides, domestic appliances, textiles, to bioremediation engineering. There are therefore concerns about the environmental risks or bioaccumulation-related issues that may arise particularly resulting from the application of drug-loaded nanocarriers or effect of pesticides that reach the natural ecosystems. This is a major threat in the present era and needs to be balanced against their undoubted benefits to human society. The assessment of the physical and chemical properties of nanoparticles and nanomaterials influencing their toxic manifestation due to accumulation in human or in animal organs is still poorly investigated. This chapter reviews the possibilities of bioaccumulation of different nanoscale particles and materials, their potential acute and subacute toxicological profile and their identification and characterization in different organs and tissues of vertebrates.

Recently, targeted drug delivery systems have gained much more interest for delivering varieties of drugs as well as imaging agents specifically to the targeted disease cells or tissues. These are well known for their increased precision and accuracy in mode of drug delivery along with reduced side effects. Though numerous carriers are being employed for drug targeting, the solid lipid based nanoparticles (SLNs) are preferred over them owing to their ability to encapsulate wide varieties of drugs, biocompatibility, ease of surface modification, scaling up feasibility, and possibilities of both active as well as passive targeting to various organs. Surface of these drug loaded SLNs can be modified by conjugating different ligands to enhance their tissue/organ targeting ability and therapeutic efficacy to much higher extent. In this chapter, we have discussed about the SLNs and their different surface modified forms for passive as well as active targeting to different organ such as (colon, breast, lungs, liver, kidney, brain, eyes, etc.) in combating different diseases.

Pharmaceutical excipients are the backbone of pharmaceutical formulations. These are inert substances added to contribute physicochemical stability in vitro as well as in vivo to the active pharmaceutical ingredients during formulation development. Excipients significantly affect the quality of a finished pharmaceutical dosage forms. They are intended to be used to facilitate the pharmacokinetic as well as pharmacodynamic profiles of the medicament. Furthermore, excipients also influence the physical properties of medicinal products including solubility, consistency, weight and volume, release rate, etc. which are required for administration of adequate dose of active drug in a particular dosage formulation. Currently, numerous synthetic excipients have been explored in pharmaceutical formulations such as tablets, capsules, novel drug delivery systems, etc. However, they are found to have some unwanted side effects in vivo after administration. Therefore, there is a quest to have some excipients from natural origin with minimum side effect or toxic manifestations.
Nature is enriched with a wide array of precious components which directly or indirectly are beneficial for maintaining the good health of living organisms. Natural excipients such as binders, diluents, sweeteners, colorants, preservatives, film formers, etc. and their derivatives are found abundantly in plants, animals, and minerals. Since natural ingredients are inert, biocompatible and biodegradable with minimum toxic effects, as well as cost-effective, hence pharmaceutical industries are showing their interest for these excipients to be used in formulating novel drug formulations, cosmetics, as well as in food products.
This book chapter entitled “Natural Excipients in Pharmaceutical Formulations” has been compiled to explore the extensive information about the excipients of natural origin to be utilized as bulking, binding, film forming, lubricating, sweetening, preservative, coloring agent, etc. in design and development of various pharmaceutical dosage forms.
Dr. Subas Chandra Dinda serving at present as Professor and Dean, School of Pharmacy, The Neotia University, West Bengal, India. His area of research interest includes design and development of Drug Delivery and Drug Targeting Systems. To date some of his research finding claimed patents and published in more than hundreds of peer reviewed journals for the benefit of scientific community.
Background
Dr. Subas Chandra Dinda born in a remote village of Nafrai (Dist: Balasore), Odisha, India and completed his early education in Sankhari High School and did D.Pharm from Orissa State Board of Pharmacy in 1992, B.Pharm from Berhampur University, 1st Class with Gold medal in 1996; M.Pharm from Andhra University with 1st Class in 1999, Ph.D. from Jadavpur University in 2008, and honored with Life Fellow from Institute of Chemists (India) in 2008.
He is found to be having wide research experience in the frontier of Drug Delivery and Drug Targeting Research covering the area of design and development of Matrix systems, Floating Drug delivery systems, Muco-adhesive microcapsules, and Nano-particle based formulations. He explored several poorly bio-available drugs through muco-adhesive as well as nano-particle based dosage forms and found to be very effective through oral route.
He also actively involved in guiding the scholars in the field of Drug Delivery System as well as interdisciplinary research covering the area of Drug Synthesis and Herbal Drug Research under the joint collaboration with the teachers from other University as well.
