Masazumi Honda was born in Mikawa Province (Aichi Prefecture), he served Ieyasu Tokugawa since he was a boy. He was the son of Masanobu.
Fought bravely in Ieyasu's many battles, including those of Sekigahara (1600) and Osaka (1614-15). After the death of Ieyasu (1616), he was in charge of construction of the mausoleum of Ieyasu in Nikko. Although he was lord of Utsunomiya Castle (Tochigi Prefecture) with a fief of 155,000 koku, he was demoted to lord of Yuri in Dewa Province (Yamagata Prefecture).
