He graduated from the Voronezh provincial gymnasium (1864).
Nikolai Alekseevich was the principal merchant in Voronezh of the beginning of the 20th century. He was a founder and head of oil pressing factory partnership «N.A. Klochkov i K» (1891), tannery partnership (1897) and other industrial enterprises.
Nikolai owned property in Voronezh and Novohopersk Districts. In 1892 played an active role in providing public assistance to victims of hunger in Voronezh Governorate. He was one of the founder members and chairman of commodities exchange (1897); councilor of the City Duma (1875-1917), city mayor (1903-1905). As a city mayor, he distinguished himself by the democratic management of the Duma meetings and desire to improve the social sphere. At the same time, he defended the interests of the largest homeowners and industrialists.
He established a number of urban facilities of social, economic and special educational profiles. Nikolai Alekseevich oversaw the construction of Nikolaev Women's Pro-Gymnasium (1897-1898), provided loans for these purposes. In 1911-1912, he provided a loan of 70,000 rubles for the construction of the city building of the secondary mechanic-technical school. He established an Integrated Municipal Programme for the development of Voronezh (1908).
He was a councilor of Voronezh provincial and district Zemstvo assemblies. He was one of the leaders of Voronezh political party organization "Union of October 17". He was one of Voronezh’s main benefactors, chairman of "Voronezh popechitelniy o bednyh komitet", treasurer and vice-chairman of "Mariinskoe popechitelstvo o slepyh", trustee of Aleksandro-Mariinskaya glaznaya lechebnitsa.
The merits of Klochkov in charity were awarded a gold medal (1893) and two orders. In 1913, Zemstvo Hospital was built near Klochkov’s estate (Rogachevka village, Voronezh district) with his funds.
The Klochkov’s House in Voronezh is preserved as a historical and architectural monument. The manor was preserved in the village of Rogachevka, Novoussmansky District.
