He graduated from the Faculty of History and Philology of the Imperial St. Petersburg University in 1885. In 1888 he defended his master's thesis.
In 1889-1895 Petukhov was a professor at the Institute of History and Philology of Nezhinsk.
In 1895-1918 worked at Yuriyev University.
He had been a member of the Russian literature Society since 1898.
Petukhov became a corresponding member of the St. Petersburg Academy of Sciences in 1916.
He was a professor at Voronezh State University in 1918-1919.
Since 1919 teached at Tavria University / Crimea State Pedagogical Institute named after Frunze.
Petukhov took an active part in resolving the issue of evacuating Yuriev University to Voronezh, where he stayed from the end of 1917 till April 1918. He headed the department of Russian language and literature at the Voronezh State University, the department of ancient books and literature at the provincial museum. He left Voronezh with White Army troops in October 1919.
